Title: Mechanisms of Host-bacterial Interactions, Ileitis and Granuloma Formation in Agr2 -/- Mice

DESCRIPTION (Provided by Applicant): The Simpson lab will assist with culturing and handling of AIEC, evaluation of colonization in vivo, and xenophagy studies of AIEC and Agr2-/- cells. My laboratory will be substantially involved in Aims 1 and 3 of the grant. Performing FISH analysis, E.coli isolation and characterization, RAPD-PCR (genetic fingerprinting), quantitative PCR for bacterial load. We will be evaluating phagoctic and epithelial cells infected with AIEC derived from AGR2 described in Aim 3. We have expertise in GFP labeling AIEC and in vitro characterization of invasion and survival. We have the ability to genetically modify AIEC and we will provide Dr Sartor with wild type and mutants of high interest based on results of concurrent, non-overlapping, studies to define virulence attributes of AIEC.
