Title: Horse Genome Project: Functional Genomics through Equine Microarrays

Description (provided by applicant): The goal of this proposal is to perform a thorough evaluation of a new large-scale (8,000 plus gene) equine microarray (Aim 1, Year 1) and to use the array (Aim 2, Year 2) in discovery research using cells and tissues from several Cornell investigators conducting immunologically related research.
Aim 1 would compare the new horse-specific array with a commercial 44,000 gene human array using control and test horse lymphocytes and trophoblast cells. The validation process would include bioinformatic comparisons and verification of expression patterns of selected genes using quantitative RT-PCR assays and immunological assays such as flow cytometry for protein expression.
Aim 2 would use the horse array to assay test and control samples from projects of five Comell equine researchers. The samples would be provided by the individual investigators, but the RNA preparation, labeling, and hybridizations would be performed by a single technician to ensure quality control. The resultant data would be returned to the investigators for use in developing new experimental approaches and projects.
This proposal will provide critical validation of an important new tool for equine researchers, and use of the array by Zweig-funded scientists before its general release.
