Title: Study of the Immunomodulatory Effect of the Hookworm Molecule Ac-TMP in the Intestinal Mucosa

DESCRIPTION (provided by applicant): This proposal aims to address the fundamental biomedical problem of inducing antigen-specific immunity to tumors by epicutaneous (skin) immunization with tumor antigen in adjuvant. This idea stems directly from my previous work in experimental autoimmune encephalomyelitis (EAE), an animal model for multiple sclerosis. In EAE, epicutaneous immunization with autoantigen alone in a skin patch inhibited the development of EAE. However, when epicutaneous immunization was performed with autoantigen in the presence of adjuvant, disease was dramatically accelerated and all mice died from EAE. T cells from mice epicutaneously immunized with autoantigen in adjuvant exhibited robust proliferation and a strong Thl cytokine profile.
