The Caddy Lab at Cornell University Baker Institute for Animal Health has established intracellular (‘AMIN’) and extracellular (‘ExN’) neutralization assays based on high-throughput fluorescent imaging. For AMIN, antibodies are electroporated into cells followed by infection with RV. For ExN, antibodies are incubated with virus prior to addition to cells. This project aims to identify the antibodies capable of mediating neutralization intracellularly, extracellularly, or via both mechanisms, and to determine which rotavirus antigens are recognized in the IgA whole virus lysate ELISA following immunization with rotavirus vaccines. The study will examine the correlation with RV-specific IgA titers using the two approaches below:
1) RV-specific monoclonal antibodies
2) Depletion of antibody subsets from polyclonal sera derived from infants immunized with either Rotarix or the TV P2-VP8 vaccine.
